SOLO si ha confirmado que su NUEVA parte compuesta (plásmido recién construido) es CORRECTA. Esto significa que después de realizar un Zymo Miniprep y digestión de restricción, el nuevo plásmido ahora contiene las partes que se han ligado entre sí.
¿Cómo hacer un stock de glicerol?
- Usando técnica estéril, alícuotas de 500 µL de glicerol al 80% en un criovial (Esto puede o no haberse hecho ya, pregúntele a su instructor)
- Etiquete el criovial claramente en la parte superior o lateral con el número de pieza, la resistencia antibiótica (amp, chlor, tet, kan), así como cualquier otro estándar de etiquetado que use su laboratorio (¡EL ETIQUETADO ES MUY IMPORTANTE!!)
- Vórtice tu cultura
- Agrega cuidadosamente alrededor de 500uL (0.5mL) de tu cultivo al criovial. Vórtice y coloque el vial sobre hielo.
Almacene el vial en un congelador -80 que haya sido designado para almacenamiento a largo plazo.
