La amplificación por PCR de la región de control mitocondrial
Hay 2 regiones hipervariables dentro de la región control de las mitocondrias. Este ejercicio amplifica solo uno de estos. Para obtener resultados más definitivos, ambos deben amplificarse y secuenciarse. Este ejercicio nos permitirá tener una idea aproximada de los orígenes de nuestra línea materna y podremos atribuirnos a diversas tribus en todo el mundo. El genoma mitocondrial humano (archivo genbank).
Primer Directo 5'-TTAACTCACCATTAGCACC-3'
Imprimación inversa 5'-GAGGATGGTGGTCAAGGGAC-3'
- PCR de las muestras de ADN previamente extraídas.
- Vierta 2% de agarosa en el aparato de fundición en refrigerador.
- Se necesitan hacer 2 geles por clase → 100ml de TBE con agarosa 2g
- Agregue 5μl de solución segura SYBR en la agarosa fundida antes de colar.
- Coloca 2 juegos de peines en el gel → en un extremo y en el medio.
- Cargar el gel con escalera de ADN y PCR.
- Ejecutar el gel a 120V durante 20 minutos.
- Visualice en el transiluminador UV.
- Documentar con una cámara para verificar la amplificación.
- El instructor presentará reacciones viables para la secuenciación.
- Analizar datos durante la sesión del Laboratorio de Bioinformática.
