    Usando las secuencias del cebador, se puede determinar el tamaño y/o ubicación de un producto de PCR. Esto se puede hacer usando BLAST o con un programa como UGENE.

    Usando BLAST

    1. Cebadores para la inserción PV92.
      • Imprimación hacia adelante: 5′ GGATCTCAGGGTGGGTGGCAATGCT 3′
      • Imprimación inversa: 5′ GAAAGGCAAGCTACCAGAAGCCAA 3′
    2. Visita BLAST: https://blast.ncbi.nlm.nih.gov/Blast.cgi?PAGE_TYPE=BlastSearch.
    3. Pega ambas imprimaciones:
      • GGATCTCAGGGTGGGTGGCAATGCTGAAAGGCAAGCTACCAGAAGCCCCAA
      • Retire los números de 5′ y 3′.
    4. Elija “Algo Similar”.
      • Localizar el lugar del producto y el tamaño.

    Usando Ugene

    1. Ejercicio usando un cebador D-Loop humano.
      • Cebador Directo 5'-TTAACTCACCATTAGCACC-3'
      • Imprimación inversa 5'-GAGGATGGTGGTCAAGGGAC-3'
    2. Descarga el archivo Genbank de muestra: Genoma Mitocondrial Humano.
    3. Abre el archivo en Ugene.
    4. Seleccione el botón “In Silico PCR” en el extremo derecho (botón de doble hélice).
      • Inserte los cebadores directo e inverso en los espacios apropiados.

    5. Se debe anotar un producto de PCR para una de las secuencias después de presionar “Buscar productos de todos modos”.
