Las condiciones de reacción para la alquilación directa del carbono alfa con LDA u otra base muy fuerte son bastante duras. Muchos compuestos orgánicos no pueden soportar el ambiente de reacción en cantidades sintéticamente útiles. Por lo tanto, Gilbert Stork, de la Universidad de Columbia, desarrolló una vía sintética alternativa. Algunas de las ventajas de usar una enamina sobre un enolato son que las enaminas son neutras, más fáciles de preparar y generalmente previenen los problemas de sobrerreacción plagados por enolatos. Como se muestra en el siguiente ejemplo, el aldehído o cetona se puede recuperar de la enamina a través de una reacción de hidrólisis.

Las enaminas actúan como nucleófilos de una manera similar a los enolatos. Debido a esto, las enaminas se pueden usar como equivalentes sintéticos como enolatos en muchas reacciones. Este proceso requiere tres etapas: 1) Formación de la enamina, 2) Reacción con un eletrofilo para formar una sal de iminio, 3) Hidrólisis de la sal de iminio para reformar el aldehído o cetona.
Normalmente utilizamos las siguientes 2 o aminas para las reacciones de enamina
Alquilación de una enamina
Los enaminados se someten a una reacción de S N 2 con halogenuros de alquilo reactivos para dar la sal de iminio. La sal de iminio puede hidrolizarse de nuevo en el carbonilo.
Pasos individuales
