Uso de Cytobrush
- Use un citocepillo estéril e inserte en la boca.
- Cepille el citocepillo en el interior de su mejilla 25 veces.
- Remolino el citocepillo en 100 μl de suspensión de Chelex (10% p/v).
- Colocar un tubo de centrífuga con Chelex y suspensión celular en un bloque térmico de 100 °C durante 10 minutos.
- Centrifugar los tubos a velocidad máxima durante 5 minutos.
- El ADN está en el sobrenadante. Evita las cuentas en la parte inferior.
- Almacenar el ADN a -20 °C.
PCR con perlas de PCR
- Añadir 22 μl de la mezcla de imprimación (hacia adelante y hacia atrás) a las perlas.
- Asegúrese de que la perla esté disuelta.
- Agregar 3 μl de ADN.
