De manera similar al aldol, un éster actúa como nucleófilo mientras que un segundo éster actúa como electrófilo. Durante la reacción se forma un nuevo enlace carbono-carbono. El producto es un β-cetoéster. Una diferencia importante con la reacción aldol es el hecho de que el hidróxido no se puede usar como base porque posiblemente podría reaccionar con el éster. En cambio, se usa una versión alcóxido del alcohol utilizado para sintetizar el éster para prevenir los productos secundarios de transesterificación.

Condensación Cruzada de Claisen
Las condensaciones de Claisen entre diferentes reactivos éster se denominan reacciones de Claisen Cruzado. Las reacciones cruzadas de Claisen en las que ambos reactivos pueden servir como donantes y aceptores generalmente dan mezclas complejas. Debido a esto, la mayoría de las reacciones de Claisen Cruzado generalmente no se realizan a menos que un reactivo no tenga hidrógenos alfa.
